Tyrosine Kinase Receptors

Tyrosine kinases are a group of enzymes that transfer phosphates from nucleotide triphosphates to amino acids. They usually get the phosphate group from ATP. The addition of the phosphate to the amino acid results in a conformational change affecting the protein function. This action is responsible for initiating many processes within a cell. Tyrosine kinase receptors are a family of receptors with a tyrosine kinase domain and a hormone binding domain. Growth factors or hormones can bind to the hormone binding domain which activates the receptor and causes them to aggregate. This aggregation causes the tyrosine kinase domain to phosphorylate certain amino acid or peptides. These phosphorylated peptides or proteins then initiate a cascade of events such as recruiting enzymes or molecules and activating G proteins. These cascades continue on to perform many important cellular functions like gene transcription.

Myofibrillar myopathy is a disease characterized by slowly progressive weakness that can involve both proximal and distal muscles. Anti-ALK antibody binds against the target anaplastic lymphoma kinase. ALK is localized on the membrane and expressed in the brain and central nervous system. Anaplastic lymphoma kinase is an orphan receptor with a tyrosine-protein kinase activity and plays a role in the normal development and function of the nervous system. Defects in ALK are found in a form of non-Hodgkin lymphoma and cause susceptibility to neuroblastoma type 3. Neuroblastoma is a malignant tumor that develops in nerve tissue. Other symptoms include bone pain, flushed skin, profuse sweating, loss of movement, problems with balance, and uncontrolled eye movements. Treatment includes radiation, chemotherapy, and possible surgery.
