Polyclonal Antibodies

Antibodies can selectively react to antigens and provide useful tools for many different applications revolving around the antibodies ability to detect a certain molecule of interest. Antibodies are made by immunizing a host animal with a foreign molecule. That animal’s B lymphocytes (or B cells) produce antibodies directed at a specific part of that foreign molecule. Each different B cell will produce its own individual type of antibody. These antibodies are considered “polyclonal” because they are made of a variety of different antibodies that each recognizes a particular site on the immunizing peptide or particle. Polyclonal antibodies are made up mainly of immunoglobulin G subclasses (IgG). Polyclonal antibodies are less expensive to produce than monoclonal, they require lower amounts of technology or skill to produce, and the time it takes for production is relatively short. Because polyclonal antibodies contain a mixture of antibodies for different aspects of the immunizing antigen, they are not useful when expecting affinity to a specific domain of an antigen. However, this does provide certain advantages. These advantages are related to their ability to bind more than one site on their target antigen. Including their ability to amplify signals from a target protein, show better results in IP/ChIP, tolerate minor conformational changes in the antigen and detect de-natured proteins. Disadvantages to polyclonal antibodies include their tendency to batch variations and possible background signals due to multiple non- specific antibodies in serum. Non- specific antibodies are produced by the host animal as part of an immune response but do not specifically bind to the particular antigen.

5HT3B antibody is the polyclonal antibody produced in goats that binds against the 5HT3B receptor. The 5HT3B receptor is one of several receptors for the neurochemical serotonin (5-hydroxytryptamine). Serotonin is a neurotransmitter and a hormone responsible for the regulation of satiety, hunger, mood, and sleep. Anti- 5HT3B antibody binds to this receptor in place of serotonin and can effectively block the binding of 5HT3B. The receptor itself is cation exclusive but otherwise is a comparatively non-selective ion channel. Defects in serotonin receptors can lead to disorders like depression, obesity, autism, and problems with sleep. Anti-cytokeratin antibody is the polyclonal antibody produced in goat that binds against the cytokeratin protein. Cytokeratin protein is involved in signal transduction of class 1 cytoskeleton intermediate filaments. The antibody can bind with numerous forms of cytokeratin including cytokeratin 4, 5, 6, 8, 10, 13, and 18. Thus, the cytokeratin antibody is rather broad spectrum. These anti-cytokeratin antibodies are specific cell markers and can be used in tumor identification and cataloging. The target protein helps to regulate kinases. Defects in cytokeratin cause several disorders including palmoplantar keratoderma non-epidermolytic (NEPPK) and congenital ichthyosiform erythroderma (BCIE). NEPPK or diffuse orthohyperkeratotic keratoderma is a genetic disorder characterized by hornlike skin growths on the palms of the hands and soles of the feet. CIE is a rare form of ichthyosis that affects skin and gives it a thickened, scaly, and flaky appearance
