Notch Signaling

The notch signaling pathway is a cell signaling system that is present in most multicellular organisms. Notch signaling links the fate of one cell to that of an immediate neighbor. It controls differentiation, proliferation, and apoptotic events in many different tissues. The dysregulation of notch pathways has been implicated in many developmental disorders and cancers. Notch proteins are actually single-pass transmembrane receptors with large extracellular domains that can regulate certain cell decisions. There are different modes of Notch action that include lateral inhibition, lineage decisions, and inductive/boundary formation. In mammals, there are four different notch receptors: NOTCH1, NOTCH2, NOTCH3, and NOTCH4. The main mechanism of action requires a ligand from outside of the cell binding to the notch receptor. This induces a proteolytic cleavage and releases an intracellular domain which can then into the cell nucleus and modify gene expression.

Anti-FOXP3 antibody detects a section of human FOXP3 protein. The FOXP3 protein is a transcription factor that controls certain immune responses and is thought to play a role in diabetes, infections (autoimmune and immunodeficiency), and anemia. Anti-NOTCH2 antibody binds against the target NOTCH2. NOTCH2 is localized on the cell membrane and in the nucleus. It is expressed in the brain, heart, kidney, lung, skeletal muscle, and liver. The notch 2 protein is a receptor on the membrane of cells and binds to ligands and anti- notch 2 antibodies. These proteins regulate the fate of the cell by influencing processes including differentiation, proliferation, and apoptosis specifically of osteoclasts or bone cells. Defects in NOTCH2 cause Alagille syndrome type 2 and Hajdu-Cheney syndrome (HJCYS). HJCYS is a skeletal disorder. Affected individuals have facial defects, osteoporosis, acro-osteolysis, and periodontal disease.
