Apolipoprotein Antibodies

Abcore has over 50 antibodies against Apolipoprotieins and their related proteins. Currently we offer antibodies which have been raised in goat, as well as some mouse monoclonal antibodies. Since we find APO research to be so important, we have tried to summarize some of the basic apolipoprotein research here, focusing on the various isoforms and diversity of the APO family including APOE, APOBEC-1, and APOL (1-6). Apolipoproteins are a family of proteins which are associated with cholesterol complexes including LDL and HDL. Historically, 4 classes of apolipoproteins (APO) are discussed, apolipoprotein A, apolipoprotein B, apolipoprotein C, and apolipoprotein D. Here, we will briefly review these lipoproteins, but also discuss APOE, APOBEC-1, and APOL (1-6).

Apilopoprotein A was one of the very first apolipoproteins to be identified and studied [1-3]. APOA1 is an exchangeable lipoprotein, along with APOC, and APOE [4]. APOA1 also self-associates, which can stabilize the APOA1 protein when it is not in complex with a lipid [5]. Apolipoprotein B (APOB) is synthesized in two major isoforms, APOB100 and APOB48. APOB48 is the more prevalent isoform [6]. APOB is responsible for cholesterol transport to peripheral tissues. Interestingly, APOB is regulated by protein degradation. APOB also is responsible for regulating VLDL complex numbers, which of course is also therefore regulated by degradation of APOB [7,8].

Apolipoprotein C-III (APOC-III) is found in a glycosylated and non-glycosylated forms. APOC-III is found in complex with both APOB complexes and HDL, and it exchanges between these 2 complexes very quickly [9]. APOC-III expression is regulated by both insulin and the insulin response element (IRE) promoter on it’s gene [10]. Increases in APOC-III expression have been associated with an increased risk of coronary artery disease, metabolic syndrome, and type 2 diabetes. [11-13].

Human apolipoprotein E (APOE) is important for a variety of neurological disorders including Alzheimer’s Disease and cerebral amyloid angiopathy. APOE is found in 3 polymorphic isoforms, APOE2, APOE3, and APOE4. Each has very small amino acid changes which change the functional properties of the individual APOE proteins [14]. APOE4 has been found to be a major Alzheimer’s disease onset and seceptability factor, and APOE2 offers protection from progression of the disesase. APOE is involved in regulation of lipid localization, and APOE receptors and APOE proteins are involved with lipolysis [15,16].

APOBEC-1 is the catalytic subunit of APOB mRNA editing enzyme. This protein is a more recent discovery, and is important for APOB organ-specific localization via APOB mRNA editing [17,18]. Finally, Apilipoprotein L (APOL) is associated with serum HDL complexes. APOL is a family consisting of six variants, which are all grouped on a very small region of the same chromosome [19]. The six variants and isoforms are APOL1 isoform a, APOl1 isoform b, APOL2, APOL3 isoform 1, APOL3 isoform 2, APOL3 isoform 3, APOL4 isoform 1, APOL isoform 2, APOL5, and APOL6. APOL1 is the only isoform which is known to be secreted.
